[177Lu]IS3010 is a preclinical, LFA-1-targeted radioligand therapeutic being developed by iSeek Biopharma as part of its ORION immuno-oncology theranostic program. It consists of a small-molecule ligand targeting lymphocyte function-associated antigen-1 (LFA-1; integrin alpha-L/beta-2) labeled with the beta-emitting radionuclide lutetium-177 (177Lu). LFA-1 is constitutively expressed on leukocytes and is overexpressed in certain hematologic cancers and immune cells within the tumor microenvironment. By targeting LFA-1, [177Lu]IS3010 delivers localized, cytotoxic radiation directly to LFA-1-expressing cells, offering a potential therapeutic approach for immuno-oncology indications. It is designed as a theranostic pair alongside the diagnostic PET imaging agent [64Cu]IS3000.
